Title: Subtelomeric Chromatin of S. pombe, a Fission Yeast

Abstract: Telomeres are crucial for protecting the genome. The key to this protective role is the specialized repressive chromatin that assembles at telomeres and subtelomeric domains. The repetitive nature of telomeric and subtelomeric sequences in many organisms, however, has hampered research efforts. Due to the relative ease of genetic manipulation and the strong conservation of crucial regulatory proteins with higher eukaryotes, the fission yeast S. pombe has served as an important model system for the dissection of chromatin biology. Mammals share a high degree of conservation for telomeres, the telomere-binding shelterin complex, and the assembly of constitutive heterochromatin at subtelomeres. Throughout this review, we aim to encapsulate recent findings that describe the assembly of various chromatin structures within subtelomeric domains in fission yeast. These include the telomere-associated sequences (TAS), the heterochromatic SH subtelomeric domains, and the ST chromatin domains that put together knob-like, dense chromatin clusters. We examine fresh information about the arrangement of subtelomeric domains, the various chromatin types that form on these sequences, and the effects of histone H3 K36 modifications on these chromatin structures. We discuss the interaction of the different chromatin subdomains and how the telomere-bound shelterin complexes and the subtelomeric domain’s own euchromatic chromatin regulators affect subtelomeric chromatin. Last but not least, we show that Set2, which regulates H3K36 methylation, is more important for telomere clustering than knob assembly within the condensed ST chromatin knob domains, which is mediated by these domains.
