Title: Tyrosine Kinase Inhibitors’ Emerging Importance in the Fight Against Cancer: How Effective Are They?
Abstract:According to GLOBOCAN 2020, there will be more than 19.3 million new cases of cancer worldwide in 2020, and 10 million people will die from the disease. Clinical manifestations demonstrated that a number of growth factor receptors with cytoplasmic and transmembrane tyrosine kinase (TK) domains are essential for the development of cancer. The prognosis and survival of patients with higher tumor grades are significantly impacted by receptor tyrosine kinases (RTKs), important mediators of numerous cellular pathways and carcinogenesis. TKIs are effective medications for the targeted treatment of various cancers. RTKs have consequently emerged as a promising therapeutic target to treat cancer. TKIs are essential mediators of signal transduction and the proliferation, angiogenesis, and apoptosis of cancer cells, according to a recent study. In this review, we explore RTKs’ key role in cancer therapy by talking about their structure and function. To date, numerous TKIs have been created, and they are very helpful in treating a variety of cancers. The therapeutic efficacy, mechanism of action, and side effects of these TKI-based anticancer drug molecules are also covered in detail. This article also focuses on TKIs that are currently being tested in clinical trials and pre-clinical research. Further, we reviewed the effects of RTK resistance on TKI clinical drugs as well as their mechanistic acquired resistance in various cancer types to gain insight into the pathophysiological mechanism of TKIs.
