Title: Cancer cells’ multidrug resistance and the critical function of P-glycoprotein
Abstract: A significant contributor to the multidrug resistance phenotype in cancer cells is P-glycoprotein (P-gp). P-gp is a protein that confers resistance and controls the ATP-dependent efflux of a variety of anticancer drugs. Numerous attempts have been made to stop P-action gp’s in order to restore the effectiveness of anticancer medications due to its broad specificity. The main objective has been to develop molecules that either compete for transport with anticancer drugs or serve as a direct P-gp inhibitor. Despite notable in vitro success, there are currently no medications that can “block” P-gp-mediated resistance in the clinic. The cause of the failure could be due to toxicity, undesirable pharmacological interactions, or a variety of pharmacokinetic issues. P-gp, on the other hand, has a big impact on the body. It guards against the entry of foreign objects and other harmful chemicals into the vital organs. Therefore, P-gp inhibitors shouldn’t interfere with the way it works in healthy cells. In-depth background knowledge in this area is required to create a P-gp inhibitor that is effective. The main goal of this review article was to outline the benefits and drawbacks of P-‘s effects on both cancer cells and healthy cells. Also mentioned were P-effects gp’s on cancer drug delivery and its role in triggering drug resistance.
