Title: Targeting Cancer and Complex Diseases: Diclofenac-Based Novel Therapeutics Development and Challenges
Abstract: Diclofenac is a non-steroidal anti-inflammatory drug (NSAID) that is frequently prescribed to treat a variety of clinical conditions. It reduces swelling, pain, fever, and aches. The prostaglandin synthesis-producing enzymes cyclooxygenase-1 and cyclooxygenase-2 are inhibited by this medication. We need new molecular systematic therapeutic approaches to lessen complex multifactorial effects in order to improve the current diclofenac-based therapies. But the main issue with diclofenac and other medications in the same class is their side effects, which include diarrhea, heartburn, kidney problems, cardiovascular problems, and signs of stomach injuries. In this article, we explain the necessity of defining the mechanisms, pharmacological characteristics, and therapeutic properties of diclofenac in order to guide future drug development against neurodegeneration and premature aging as well as to enhance cancer treatment. Additionally, we discuss a number of cutting-edge molecular mechanisms and fundamental aspects associated with diclofenac that can help to improve the design of new derivatives of the drug and help them overcome difficult problems and expand their applications.
