Title: Patients with diffuse large B cell lymphoma exhibit Sp17 protein expression and Major Histocompatibility Class I and II epitope presentation.
Abstract: Patients with diffuse large B cell lymphoma urgently require better treatments (DLBCL). The development of chimeric antigen receptor T cells and immune checkpoint inhibitors has increased the need for validated cancer epitopes. With their widespread expression in many tumors but extremely constrained distribution in normal tissue, immunogenic cancer testis antigens (CTAs) represent appealing immunotherapeutic targets that may enhance the available therapies for DLBCL and other malignancies. Sperm protein 17 (Sp17), a CTA that has been linked to myeloma and ovarian cancer, is expressed in DLBCL. The current study’s objective was to examine Sp17 epitope presentation by looking at CD4 T-helper (Th) and cytotoxic T cell (CTL) responses in patients with DLBCL. Following brief cell stimulation with two novel HLA-A-0201 peptides and one previously reported HLA-A-0101-restricted nine-mer Sp17 peptide, a significant -interferon CTL response was found in peripheral blood mononuclear cells of 13/31 DLBCL patients. Both the four healthy subjects and the HLA-A-0201-negative DLBCL patients showed no discernible responses. In 8/17 DLBCL patients, a brand-new immunogenic 20-mer CD4ThSp17 peptide was found. This is the first account of a CTL and CD4 response to Sp17 in a DLBCL patient, supporting Sp17 as a potential immunotherapeutic target.
