Title: The screening of four Hypericum species from the Canary Islands revealed their bioactive potential.
Abstract: In this study, we offer a general methodology to evaluate extracts’ bioactive potential (BP) in the search for plant-based medicines. In order to demonstrate the methodology, we looked at the anticancer potential (AP) of four endemic Hypericum species from the Canary Islands: Hypericum canariense L, Hypericum glandulosum Aiton, Hypericum grandifolium Choisy, and Hypericum reflexum L.f. Six human tumor cell lines—A549 (non-small-cell lung), HBL-100 (breast), HeLa (cervix), SW1573 (non-small-cell lung), T-47D (breast), and WiDr—were tested using microextracts made from the aerial parts of these species (colon). Cell migration, autophagy, and cell death were also examined in more detail in the methanol-water microextracts. By using UHPLC-DAD-MS, the most promising bioactive polar microextracts were examined. The bioactivity assessment, extraction yield, and chemical profiling by LC-MS all indicated that H. grandifolium had the highest AP among the species tested. HeLa cells exposed to a methanol-water microextract of H. grandifolium allowed for label-free live-cell imaging studies that allowed the observation of cell death and several apoptotic hallmarks. Overall, this research enables us to choose Hypericum grandifolium Choisy as a source of novel chemical compounds that may be useful in the treatment of cancer.
