Title: Auricularia auricula Polysaccharides’ Effects on Type 2 Diabetic Mice’s Gut Microbiota Composition
Abstract: Auricularia auricula polysaccharides (AAP) have been shown in prior studies to improve type 2 diabetes mellitus, but its exact mechanism is still unknown. In this study, we aimed to show that AAP induces remission in type 2 diabetic mice by modifying the gut microbiota. After successfully creating a type 2 diabetes mellitus (T2DM) model using a high-fat diet and streptozotocin (STZ), we saw significant reductions in fasting blood glucose (FBG) levels and oral glucose tolerance test (OTGG) after 5 weeks of AAP intervention. To further mitigate the oxidative stress injury, AAP increased the activities of total superoxide dismutase (T-SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) and decreased the concentration of malondialdehyde (MDA). The best improvement effect was seen with AAP-M (200 mg/kg/d). Additionally, findings from 16S rRNA indicated that AAP increased Bacteroidetes abundance while decreasing Firmicutes abundance. The consumption of AAP led to an increase in the diversity of advantageous genera like Faecalibaculum, Dubosiella, Alloprevotella, and those in the family Lachnospiraceae. Desulfovibrio, Enterorhabdus, and Helicobacter could have less of a population due to AAP. These findings collectively imply that AAP can treat disorders of lipid and glucose metabolism by regulating the composition of the gut microbiota.
