Title: Through the Nicholas Reaction, functionalized 10-membered aza- and oxaenediynes are created.
Abstract: In order to create 10-membered benzothiophene-fused heterocyclic enediynes with various functionalities, the range and restrictions of the Nicholas-type cyclization were examined. Although an ester group allowed for the Nicholas cyclization through oxygen to occur, the resulting oxaenediyne was unstable when stored. The N-type Nicholas reactions that produce 10-membered azaendiynes in high overall yields include cyclization via an arenesulfonamide functional group followed by mild Co-deprotection. The desired 10-membered cycle was not produced by the Nicholas cyclization through the acylated nitrogen atom, however. It led to the formation of a pyrroline ring, as opposed to the poor yield of the desired 10-membered enediyne that was produced by cyclization via an alkylated amino group. The most practical nucleophilic group for the synthesis of functionalized 10-membered enediynes with a clickable function, such as a terminal triple bond, was discovered to be the acylated 4-aminobenzenesulfonamide nucleophilic group. All of the synthesized cyclic enediynes had a low to moderate effect on lung epithelial-like WI-26 VA4 cells and exhibited moderate activity against lung carcinoma NCI-H460 cells, making them promising compounds in the search for new antitumor agents that can be combined with tumor-targeting ligands.
