Title: Streptozotocin-Induced Type I Diabetes-Associated Reproductive System Dysfunction and Inflammation: Protective Effect of Portulaca oleracea
Abstract: Background: Type 1 diabetes (T1D), an autoimmune disease with persistent inflammation, has implications and complications for infertility. This study sought to determine the protective effect of Portulaca oleracea (PO) plant extract on streptozotocin-induced type I diabetes-associated reproductive system dysfunction and inflammation. PO has been shown to have antidiabetic, antioxidant, and anti-hyperlipidemic properties. Methods: Four experimental groups of male rats were created at random: control, diabetic, and treatment (PO extract at 100 or 300 mg/kg/day). Then, the subjects’ intake of food and liquids, body, testis, and epididymis weights, histopathological analysis, diameter of the seminiferous tubules, sperm count, and motility, glucose levels, sex hormones, and markers of oxidative and inflammatory stress were assessed. Results: The consumption of food and liquids increased significantly in streptozotocin-induced diabetes, as did glucose, MDA, TGF-1, and TNF- levels. Conversely, the diameter of the seminiferous tubules, sperm count, and motility, as well as the levels of LH, testosterone, total thiol, VEGF, and SOD activity, decreased. It’s interesting to note that PO extract significantly reduced the damage to these parameters and histopathological indices in rats. PO extract was phytochemically characterized by using liquid chromatography-mass spectrometry to detect bioactive molecules. Conclusion. The findings of this study will open the door to the rational application of PO on clinical complications and implications associated with diabetes, even though more preclinical evaluations are required to more fully characterize the mechanism(s) of action.
