Title: Salvianic Acid A Synthesis from L-DOPA Using Biocatalytic Cascade Reactions

Abstract: The traditional Chinese herb Salvia miltiorrhiza’s primary bioactive component, salvianic acid A (SAA), has significant therapeutic potential for the treatment of cardiovascular diseases. In this study, a two-step bioprocess was created for producing SAA from L-DOPA. In the initial step, membrane-bound L-amino acid deaminase from Proteus vulgaris was expressed in engineered Escherichia coli cells to convert L-DOPA to 3,4-dihydroxyphenylalanine (DHPPA). Then, permeabilized recombinant E. coli cells co-expressing D-lactate dehydrogenase from Pediococcus acidilactici and formate dehydrogenase from Mycobacterium vaccae N10 converted the unpurified DHPPA directly into SAA. With a 96.6% yield and optimal conditions, 48.3 mM of SAA could be produced from 50 mM of L-DOPA. The bioprocess created here is therefore promising for the production of industrial SAA because it was not only environmentally friendly but also showed excellent production efficiency.
