Title: Origanum majorana L. Essential Oil from Nepal: Chemical Composition, Enantiomeric Distribution, Antimicrobial and Antioxidant Activities

Abstract: The objective of this study was to investigate the chemical composition, biological activities, antioxidant capacities, and enantiomeric compositions of Nepalese-origin Origanum majorana L. essential oils (EOs). Using a Clevenger-style apparatus, the EOs were hydro-distilled to extract them, and gas chromatography and mass spectrometry were used to determine the chemical compositions of the extracted EOs (GC-MS). To assess the enantiomeric compositions of EOs, chiral GC-MS was employed. The 2,2-diphenyl-1-picrylhydrazyl scavenging assay and ferric-reducing antioxidant power were used to measure the antioxidant activity and determine the minimum inhibitory concentrations (MICs) of the essential oils (FRAP). The EO samples (S1) and (S2), which represent the Kathmandu and Bhaktapur districts, contained 50 and 41 compounds, respectively, according to GC-MS analysis. Terpinen-4-ol and the oxygenated monoterpenoids made up the majority of the two EO samples. The major components of the EOs from the two locations did, however, differ slightly from one another. According to the literature review, the chiral terpenoids for two EO samples of marjoram have also been elaborately reported in this study for the first time. There are at least 5 distinct chemotypes of marjoram oil, according to a hierarchical cluster analysis based on the compositions of EOs with 50 compositions reported in the literature. In comparison to the used standard, the antioxidant activity for the sample (S2) was found to be relatively moderate, with an IC50 value of 225.61 0.05 g/mL and an EC50 value of 372.72 0.84 g/mL. Additionally, the EO from sample (S2) showed potent antifungal activity against Candida albicans and Aspergillus niger with a MIC value of 78.1 g/mL. Additionally, both samples showed significant antimicrobial activity. The findings imply that EOs of Origanum majorana have notable antimicrobial and antioxidant activity, making them suitable for use as a natural preservative in the food and pharmaceutical industries.
