Title: Recent Advances in Drug Discovery: Molecular Insights into Coumarin Analogues as Antimicrobial Agents

Abstract: The emergence of pathogens that are multidrug resistant and drug-resistant bacteria that are associated with high mortality pose a serious threat to global health. Different plants are rich in coumarins, which are heterocyclic benzophenone class compounds. Numerous biological activities, such as those that are antibacterial, anticoagulant, antioxidant, anti-inflammatory, antiviral, antitumor, and enzyme inhibitory, are exhibited by coumarins and their derivatives. The optimization, synthesis, and evaluation of novel coumarin analogues as antimicrobial agents have all been reported in recent years. The majority of the coumarin-based antibiotic hybrids that have been developed have been reported to show potential antibacterial effects. The focus of the current work is on studies about antimicrobial coumarin analogues that were published between 2016 and 2020. The capacity of coumarins to scavenge free radicals accounts for their wide range of biological properties. Some of the structural characteristics include a heterocyclic ring with electron-withdrawing/donating groups conjugated with the coumarin nucleus, in addition to the various synthetic techniques developed. The suggested structure–activity relationship (SAR) can shed light on how coumarin hybrids can be logically improved to combat bacteria with a wide range of drug resistance. The current work illustrates molecular understanding for recent coumarin derivatives with antimicrobial properties. The comprehensive SAR results will help guide optimization efforts during the development of new antimicrobial therapeutics.
