Title: Histone deacetylase inhibitors as a Novel Epigenetic Therapy for Bacterial Infections: Increasing Cationic Antimicrobial Peptide Production
Abstract: The development of antibiotic resistance poses a serious threat to our ability to effectively treat a variety of common and crucial bacterial infections. We require novel therapeutics that can be used either independently or in combination with antibiotics. The crucial host innate defense agents known as cationic antimicrobial peptides (CAMPs) have broad-spectrum activity against a variety of microorganisms. Numerous cell types, including epithelial cells, express CAMPs either constitutively or under the influence. Phagocytic granules carry CAMPs. The ability of HDAC inhibitors (HDACi) to prevent the action of HDACs to increase CAMP expression as well as the role of histone modification enzymes, specifically the histone deacetylases (HDAC), in down-regulating the transcription of CAMP-encoding genes are both becoming more widely recognized. Therefore, increasing CAMPs on mucosal surfaces using artificial and organic HDACi molecules may have therapeutic benefits. Here, we examine how the induction of HDACs by both the host and the pathogen regulates the expression of CAMP, and we evaluate the therapeutic potential of both natural and synthetic HDACi using data from tissue culture systems, animal models, and clinical trials.
