Title: Novel treatments for malignant pleural mesothelioma open up new possibilities.
Abstract: Malignant pleural mesothelioma (MPM), a cancer of the pleural mesothelial cells, is comparatively uncommon but extremely deadly. Exposure to asbestos is a key factor in its pathogenesis. Despite recent advancements offering a more thorough understanding of the pathogenesis, the results remain dismal. The gold standard of treatment is still trimodality therapy, which combines surgery with chemotherapy and/or radiotherapy. The management of this cancer is made more difficult by the tumor cells’ emergence of resistance to radiation and a number of chemotherapeutic agents. Ionizing radiation causes DNA damage to cancer cells and promotes therapeutic response, but it also turns on cell survival signaling pathways that enable tumor cells to withstand the cytotoxicity brought on by radiation. A thorough analysis of the biology underlying mesothelioma, with a focus on how pro-survival signaling pathways function, may provide insight into potential treatment options. This review focuses on the current MPM treatment options as well as novel treatment strategies based on recent studies that combine radiotherapy with inhibitors that target various pro-survival pathways.
