Title: Collisions between transcription and replication—A Series of Unlucky Occurrences
Abstract: When DNA replication comes into contact with genomic regions undergoing transcription, transcription-replication interactions take place. Conflicts between replication and transcription—both of which use the same DNA template and are necessary for life—are inevitable. Processive replication or transcription may be hampered by R-loops, DNA supercoiling, DNA secondary structure, and chromatin-binding proteins. When these processes co-occur, the threat to genome integrity is even greater. The importance of defining cellular pathways regulating transcription-replication interaction formation, evasion, and resolution is highlighted by the necessity of maintaining high fidelity and processivity of transcription and replication while navigating through a complex chromatin environment. In this article, we’ll talk about how transcription affects replication fork stability and the safeguards that have developed to help mammalian cells navigate transcription-replication interactions and preserve genome integrity.
