Title: A Justification for PAR1-Targeting Therapies: The Evolving Concept of Neuro-Thrombo inflammation for Neurodegenerative Disorders and Neurotrauma
Abstract: Before the functional thrombin receptor was cloned in 1991, several laboratories were actively interested in the function of coagulation and fibrinolysis in the nervous system. One of those was the role of thrombin and plasminogen activators in the development and destruction of synapses in the neuromuscular system, with a focus on diseases like amyotrophic lateral sclerosis (ALS) and how clotting and fibrinolytic pathways contribute to its pathogenesis. This viewpoint emphasizes the emerging idea of neuro-thromboinflammation that has been the subject of studies and reports for more than three decades. It highlights how, with a focus on PAR1, ALS, and parmodulins, it may result in novel therapeutic approaches to treat the effects of neurotrauma and neurodegenerative diseases.
