Title: The role of adipocyte triglyceride lipase in hepatic pathophysiology

Abstract: Triglycerides (TG) are actively oxidized by the liver to produce energy. Toxic lipid buildup, oxidative stress, and ultimately liver damage are all symptoms of metabolic disorders in the liver brought on by an imbalance between TG synthesis and hydrolysis. The enzyme that catalyzes the initial step of the breakdown of TG into glycerol and fatty acids is known as adipose triglyceride lipase (ATGL). Although the adipose tissue, heart, and skeletal muscle where ATGL regulates lipid homeostasis have been relatively well studied, it is still unclear how and to what extent ATGL is controlled in the liver, reacts to stimuli and regulators, and mediates disease progression. The current understanding of the structure-function relationship of ATGL, the molecular mechanisms of ATGL regulation at translational and post-translational levels, and most importantly—its role in lipid and glucose homeostasis in health and disease with a focus on the liver—are discussed in this review. The development of targeted therapies for the treatment of hepatic metabolic disorders depends on advances in our understanding of the molecular mechanisms underlying hepatic lipid accumulation.
