Abstract: Treatments for cancer may be more successful if the metabolic limitations of a tumor are better understood. Evidence supporting the crucial aspartate dependency of many tumor types has surfaced in recent years. Aspartate is necessary for cell growth because it is a precursor for the synthesis of nucleotides. Additionally, the malate-aspartate shuttle is important for maintaining redox balance, and an aspartate deficiency can result in oxidative stress. It is now understood that respiration and glutaminolysis in cancer cells, as well as mitochondrial metabolism, control a large portion of aspartate biosynthesis. As a result, aspartate can become a limiting factor for tumor growth and cancer cell survival under circumstances that suppress mitochondrial metabolism, such as mutations, hypoxia, or chemical inhibitors. Notably, the availability of aspartate has been linked to sensitivity or resistance to a variety of therapeutics that are currently being used in clinical trials or in the clinic, supporting the urgent need for more efficient aspartate-targeting strategies. In this review, we outline the most recent findings regarding the metabolic functions of aspartate in cancer cells and explain how these cells maintain aspartate levels in various metabolic conditions. We also list a number of promising aspartate level-modulating compounds that are being researched right now.
