Title: Using Cryo-EM Data Heterogeneity to Examine Biomolecule Structural Modification
Abstract: Cryo-EM, or single-particle cryogenic electron microscopy, is now a vital tool for examining the fine structural details of biomolecules. It allows for the direct visualization of the biomolecules and creates the possibility of reconstructing a three-dimensional Coulomb potential density map by averaging molecular images. Newly created data analysis algorithms enable the extraction of structural heterogeneity from a large and low signal-to-noise-ratio (SNR) cryo-EM dataset, enhancing our comprehension of the target biomolecule’s multiple conformational states or additional implications in dynamics. The workflow of single-particle cryo-EM, including imaging and data processing, as well as newly developed techniques for analyzing data heterogeneity to comprehend the structural variability of biomolecules, are briefly described in this review’s overview.
